S665 : Effect of the cold plasma on the fungal spores contaminating the spawm of edible mushrooms

Abstarct: Abstract The purpose of this study was to investigate the effects of sodium hypochlorite (Bleach), atmospheric cold plasma by surface dielectric barrier discharge method and also their synergistic effect on the inactivation of spores of two fungal species, Aspergillus flavus and Aspergillus tubingensis, which contaminate the seeds of edible and medicinal mushrooms. In the sodium hypochlorite treatment, the spore samples were exposed to different concentrations of bleach at different hours, and in the cold atmospheric plasma treatment, they were exposed to plasma radiation at different times and powers. Finally, in order to identify the optimal conditions in terms of energy consumption, the combined treatment of sodium hypochlorite and cold atmospheric plasma was performed and the results were reported as the number of colony forming units (CFU) in each sample. The field emission scanning electron microscope images showed the disintegration of the spore structure and the reduction of their size compared to the control samples. The results of X-ray energy diffraction spectroscopy showed that the most abundant element in the control samples is carbon and the amount of this element decreased in the treated samples; while the level of oxygen and nitrogen increased. The epifluorescence microscope images showed the resistant structure of the cell wall of the spores in the control samples, which preveanted staining with fluorescent dyes to some extent; While the cell wall of the spores was destroyed in the treated samples, and this led to color acceptance.
